野生型およびHis^<61>をPheもしくはSerに置換したP-糖タンパク質(PGP)を発現したLLC-PK1を作出した。これらのPGP発現細胞のvinblastineおよびcolchicine耐性は増強し,細胞内幕積は減少していた。PCB-126はフィルター上に培養したPGP発現細胞でvinblastineの経上皮細胞層輸送を阻害した。 |

The LLC-PK1 cells expressing wild type P-glycoprotein (POP) and mutant POP in which His^<61> was substituted with Phe or Ser were prepared. In these POP expressing cells, tolerances to vinblastine and colchicine were increased, and accumulations of these drugs were reduced. PCB-126 inhibited transepithelial transport of vinblastine in these PGP expressing cells on the bottom filtered well. |
