| 内容記述 | 胎子動脈管の母体indomethacin投与による収縮から再び拡張するまでの回復過程におけるNOの関与を検討するために，indomethacinによる収縮が最も強く発現するindomethacin (1 and 10 mg/kg) 投与後3時間にNO合成酵素 (NOS) 阻害剤であるN(G)-nitro-L-arginine methyl ester (L-NAME; 50 mg/kg) を投与しその後の動脈管の変化を観察した。 妊娠20日の母ラットにindomethacinを投与し，その3時間後にL-NAMEを投与すると，その子の動脈管はindomethacinの投与後24時間 (L-NAMEの投与後21時間) において，対照群と比較して有意な低値を示し，動脈管は収縮していた。観察された胎子動脈管の収縮の程度は，indomethacin投与後3時間でみられた収縮とほぼ同等であり，indomethacinによる収縮が持続し再拡張過程にはないことが明らかとなった。 以上のとおり，胎子動脈管の母体indomethacin投与による収縮から再び拡張するまでの回復過程におおいてNOのが重要な役割を担っていることが示唆された。 |

| 内容記述 | We studied the effects of N(G)-nitro-L-arginine methyl ester (L-NAME), a nitric oxide (NO) synthase inhibitor, on the ductus arteriosus (DA) that constricted by maternal indomethacin administration in fetal rat. Pregnant rats were administered indomethacin (1 and 10 mg/kg) orally 24 hr before cesarean section, and then L-NAME (50 mg/kg) was injected intraperitoneally 3 hr after indomethacin administration. The caliber of the fetal DA was studied with a whole-body freezing method. Indomethacin alone group showed that the fetal DA was reopened 24 h after maternal indomethacin administration. However, compared with the indomethacin alone group, L-NAME-treated group showed a significant constriction of the DA 24 h after maternal indomethacin administration. These results indicate that NO has a significant role in the course of reopening of the fetal DA that constricted by maternal indomethacin administration. |
