{"created":"2023-06-19T07:19:07.932530+00:00","id":5032,"links":{},"metadata":{"_buckets":{"deposit":"1a6535a6-bb5a-4ea3-81c1-1895a3a33a17"},"_deposit":{"created_by":4,"id":"5032","owners":[4],"pid":{"revision_id":0,"type":"depid","value":"5032"},"status":"published"},"_oai":{"id":"oai:az.repo.nii.ac.jp:00005032","sets":["17:40:280","31:181:494"]},"author_link":["21777","21781","21778","21780","21782","21779"],"item_12_biblio_info_16":{"attribute_name":"書誌情報","attribute_value_mlt":[{"bibliographicIssueDates":{"bibliographicIssueDate":"2008-03-31","bibliographicIssueDateType":"Issued"},"bibliographicPageEnd":"133","bibliographicPageStart":"130","bibliographicVolumeNumber":"15/16","bibliographic_titles":[{"bibliographic_title":"麻布大学雑誌"},{"bibliographic_title":"Journal of Azabu University","bibliographic_titleLang":"en"}]}]},"item_12_description_13":{"attribute_name":"抄録","attribute_value_mlt":[{"subitem_description":"TGF-βは,細胞の分化・増殖を調節する分泌性蛋白質である。TGF-βはII型受容体に結合した後,I型受容体(Alk5)と会合し,情報を細胞内に伝達する。ラット血管平滑筋細胞では,細胞外領域をコードする12塩基の選択的スプライシングの結果生じる,2つのAlk5 isoform-short form: Alk5-Sおよびlong form: Alk5-L-の存在が知られているものの不明な点が多い。本研究は,マウス組織ならびに各種細胞株におけるAlk5 isoformの遺伝子発現ならびにAlk5 isoformを介した情報伝達を検討した。マウスの臓器(肝臓,腎臓,脾臓,心臓,肺,脳,精巣,筋肉)におけるAlk5 isoformの発現をRT-PCRならびにPAGEにより比較したところ,Alk5-SとAlk5-Lは調べた臓器全てにおいて発現が見られ,発現比は臓器間で違いはなかった。また,ES細胞様細胞(P19),色素細胞(B16),マスト細胞(P-815, BMMC)とマクロファージ(Raw264)においてもAlk5 isoformの発現が見られ,発現比に違いはなかった。次に, Alk5 isoform間での転写活性能を比較した。Alk5を介して情報伝達を行うリガンドとして,TGF-βだけでなく,GDF-8も知られている。そこで,TGF-β_1ならびにGDF-8の情報伝達に関して,機能的なAlk5を欠損しているL17細胞を用いて検討した。Alk5の活性化によりFoxH2依存的に転写が促進されるAR3-lucをreporterとしてluciferase-based reporter assayを行ったところ,いずれのAlk5 isoformを発現させた場合でも,TGF-β_1とGDF-8はともに用量依存的にAR3-lucの転写を促進した。Alk5発現時の各リガンドに対するEC_<50>濃度は,TGF-β_1をリガンドとした場合もGDF-8をリガンドとした場合もAlk5 isoform間で違いはなかった。以上のことから,Alk5の2つのアイソフォームはマウスの様々な臓器および細胞で発現しており,今回調べた活性に関して,これらの両方が同等にTGF-β/GDF-8の受容体として機能していると考えられた。","subitem_description_type":"Abstract"}]},"item_12_description_14":{"attribute_name":"Abstract","attribute_value_mlt":[{"subitem_description":"Mouse TGF-β type I receptor, Alk5, encodes two types of mRNA generated by alternative splicing of four amino acids in the extracellular region. In the present study, the expression and function of alternative splice variants of Alk5, i.e., Alk5-L (Alk5 with the four amino acids) and Alk5-S (Alk5 without the four amino acids), were examined. Expression of Alk5 was detected in all examined tissues, and no significant differences in the ratio of Alk5-S to Alk5-L were detected between tissues. Expression of Alk5 was also detected in various cells, with Alk5-L being especially abundant in A-6 ES cells. No clear difference in Smad2 phosphorylation in response to treatment with TGF-β_1 was detected between L17 cells expressing Alk5-S and those expressing Alk5-L. Consistent with these results, transcription mediated by Alk5-S in L17 cells treated with TGF-β_1 was comparable to that mediated by Alk5-L. In addition, alternative splicing had no effect on transcriptional activation induced by GDF-8, a member of the TGF-β family.
