 note = {動脈管の胎生期における拡張維持と出生後の収縮閉鎖過程においてプロスタグランジン(PG) E_2が大きく関与していることは広く知られている。PGE_2の受容体は,EP_1,EP_2,EP_3及びEP_4の4つのサブタイプの存在しているが,ラットにおいて周生期動脈管におけるこれらの各PGE_2受容体の発現動態に関する報告は未だなされていない。そこで本研究は,出生前後のラット動脈管における4種のPGE_2受容体の発現動態をRT-PCR法を用いて明らかにするとともにPGE_2受容体選択的アゴニストを母体に投与しその子の動脈管の変化を観察した。その結果,ラット胎子及び新生子動脈管において,すべてのEP_1,EP_2,EP_3及びEP_4受容体が発現していることが明らかとなった。一方,EP_2受容体選択的作動薬の母ラットへの投与は,その子の動脈管に何ら影響を及ぼさなかった。, The expression of prostaglandin (PG) E_2 receptor subtypes in the ductus arteriosus (DA) was studied in the perinatal rat. In addition, the effects of EP_2 receptor selective agonist on the fetal DA were also examined. In the first series of experiments, EP_1, EP_2, EP_3 and EP_4 receptor mRNA expressions were investigated in the DA in fetal (day 21 of gestation) and newborn (1 hour and 3 hours after delivery) rats by RT-PCR analysis. In the second series of experiments, pregnant rats were given intravenous injections of EP_2 receptor selective agonist (L-lysine (z)-7-[(1R, 2R, 3R, 5R)-5-chloro-3-hydroxy-2-[(E)-(S)-4-(1-ethylcyclobutyl)-4-hydroxy-l-butenyl] cyclopentyl]-5-heptenate) at 3, 10, and 30mg/kg. The DA and pulmonary artery (PA) in the fetal rats were calibrated using a whole-body freezing method on day 21 of gestation. The results were follows : (1) all PGE_2 receptor subtypes were expressed in the DA in fetal and newborn rats. (2) The DA caliber and the DA/PA ratio of the EP_2 receptor selective agonist-treated group were not significantly different from those in the control group. It was concluded that all PGE_2 receptor subtypes were expressed in the DA in both fetal and newborn rats and that EP_2 receptor selective agonist has no constrictive or dilating effects on the DA in fetal rats., P(論文), 特集, application/pdf, FEATURE ARTICLES},
