Regulation of cell division is a crucial aspect in the development of multicellular organisms. The embryonic gonad in Drosophila carries about 12 primordial germ cells (PGCs). PGCs proliferate in the female gonad during the subsequent three larval stages until, at the late-third larval stage, the gonad carries over 100 germ cells. Here we show that profilin gene is required for proper mitotic division of PGCs during larval growth. Profilin mutant contains a reduced number of PGCs that are occasionally binucleated. Inverse PCR revealed that the insertion site of our P-element insertion allele is within the intron 1. Three ligands for profilin have been well-characterized in vitro: actin monomers, membrane polyphosphoinositides and poly-L-proline.
