Improvement of the target detection accuracy by RT-PCR is very important for molecular biological approaches. FCV has been reported to be divided into 2 groups, GI and GII, based on the capsid region (bases 6419-7053). We performed RT-PCR using Seal-S/Seal-A primers, as used for grouping in previous reports, and identified the presence of undetectable field strains. To widely detect field isolates using the capsid region, we prepared new primers consisting of sequences outside the reported primer sequences, and investigated the detection accuracy. Eight (including 3 strains isolated from vaccinated cats) of 30 FCV strains (27.0 %) were not detectable using Seal-S/Seal-A primers. The detection rates with the new primers were: 8F-S/8R-A, 100 %; AO-S/AO-A, 100 %; and CP-S/CP-A, 96.7 %. Undetectable strains had marked mutations in the primer binding sites. The mutation rate of the Seal-S binding site was 29.2 %, whereas the rate was 6.9 % in detectable strains. Seven of the 8 undetectable strains belonged to GII, which consists of only Japanese strains, showing a regional influence. Since the undetectable stains included 3 isolates (37.5 %) from vaccinated cats, the presence of such vaccine breakdown strains with marked antigen mutation in vaccinated cats should be considered in clinical diagnosis by PCR.
